MeltonVEC™-Cell Fusion Kit (HVJ-E)(Freeze dried)

No.MSV-1002
MeltonVEC™-Cell Fusion Kit is a highly effective cell fusion reagent product that uses the Sendai virus envelope (Hemagglutinating virus of Japan Envelope; HVJ-E) to inactivate the Sendai virus RNA genome, resulting in non-proliferative and non-infectious vesicle HVJ-E. It can be used in biosafety level 1 (BSL1) laboratories.Cell fusion efficiency exceeds 80%.

Nuclear-transfer Test in International Center for Primate Brain Research(ICPBR) ,Shanghai city, China

Product Application Areas:
  • The preparation of hybridoma-HVJ-E cell fusion reagent promotes cell membrane fusion by binding its envelope protein with the host cell membrane, resulting in the formation of hybridoma cells with the ability to produce monoclonal antibodies and unlimited proliferation potential.

  • It is applied in development, reproduction, and breeding research, such as nuclear transfer in enucleated unfertilized eggs—primarily by promoting the fusion of the fertilized egg with the membrane of the cell providing the nucleus through its envelope protein, allowing the recipient egg cell to accept the donor cell's nucleus, thereby achieving nuclear transfer and subsequent embryo development and species reproduction.
  • Spindle Transfer: These reagents facilitate spindle transfer procedures, a technique commonly used in reproductive biology to transfer the spindle apparatus between oocytes, supporting successful cell fusion and genetic manipulation. (The recommended soaking time in spindle transplantation experiments should not exceed 5 seconds.)

  • Oncolytic Virotherapy: In cancer treatment, cell fusion reagents assist in the development of oncolytic viruses, which selectively target and destroy cancer cells, representing a promising therapeutic approach.(Mast cell tumors, MCTs)
  • VLP(Gene Delivery Vector): Plasmids, decoy nucleic acids (DNA), and small interfering RNA (siRNA), as well as messenger RNA (mRNA), are macromolecules that require a delivery system called a "vector" to be efficiently introduced into target tissues or cells. HVJ-envelope (HVJ-E) uses the virus’s membrane fusion mechanism, allowing for the active delivery of DNA or RNA into target cells in a manner similar to injection. Unlike passive uptake via phagocytosis, this method involves the vector itself fusing with the cell membrane, offering a more efficient delivery. While utilizing the virus’s membrane fusion mechanism, HVJ-E does not use the viral genome, making it a non-viral hybrid vector. Initially developed for gene and nucleic acid delivery, it has now progressed to clinical trials as a therapeutic for cancer and infectious diseases, attracting attention as a novel gene nucleic acid delivery vector.

Product Usage Instructions

1. Upon receiving the product, please immediately check the dry ice inside the insulated container. Proper low-temperature storage is critical to maintaining the product's performance and may affect cell fusion efficiency.(We recommend that liquid nitrogen be used for quick freezing immediately after packaging to maintain activity)

2. The lyophilized powder can be dissolved using PBS (calcium-free), the amount of PBS added to the virus lyophilized powder is the volume recommended by the CoA (the batch mailed to you should be dissolved in 210ul). The virus solution dissolved in PBS does not need to be diluted and can be operated using the original solution.

3. Contact time between virus and cells After testing, other customers believe that 30s-45s is more appropriate (monkey cells), but too long contact time between virus and cells may cause cell death. The contact time between virus and cells may need to be optimized to achieve the best experimental results.
